The following seminar will be of interest to anyone using microscopy
on live cells. Bob Clegg is one of the founders of FLIM and has
continuously been involved in developing new advances and applications
in FLIM
Dr Bob Clegg, University of Illinois, Urbana-Champaign, USA, will give
a seminar in the WTB Seminar Rooms at 2.00 pm
"Fluorescence lifetime imaging (FLI) - How it helps us excavate the
rugged multi-spectral fluorescence terrain of complex biological
systems."

Abstract:
Fluorescence is a very powerful method for sensitive and detailed
analysis of molecular mechanisms. One of the primary goals of physical
molecular biology is to understand biological mechanisms on the
molecular scale, and fluorescence imaging is a powerful technique for
analyzing cellular biological systems. However, in a complex
environment of a biological cell, or living tissue, the situation
rapidly becomes exponentially more difficult than in a simple
cuvette-based system to derive quantitative data. These difficulties
pose a fascinating challenge for fluorescence analysis. This talk will
describe fluorescence lifetime imaging, which is a method of acquiring
fluorescence lifetimes at every pixel of a fluorescence image. A short
introduction will describe our instrumentation and review the major
reasons why we are interested in fluorescence lifetimes what type of
information is available from the fluorescence lifetimes. Then several
applications of FLI will be presented, including studies of
photosynthesis using the live algae Chlamydomonas, diagnosis of
prostate cancer biopsies, studies of ALA instigated protoporphyrin IX
production in living cells, pH variation across human skin, and recent
results on a new redox sensor. In addition, some new methods of
analysis will be presented that increase dramatically the ability for
analyzing FLI data from complex living systems.
